A metabolic profile of all-cause mortality risk identified in an observational study of 44,168 individuals
14 blood biomarkers predicted mortality better than conventional risk factors.
Longevity Clock is developing a blood test that measures 80+ metabolites and turns them into a biological age estimate for six organ systems: liver, kidney, cardiovascular, neuro, mitochondrial and metabolic. It is a wellness tool, built on published cohort research, with calibration for Indian and European populations as a core goal.
Organs don't age in lockstep. In a study of 5,676 adults, nearly 1 in 5 people showed strongly accelerated aging in at least one organ.
Oh et al., Nature 2023 · doi:10.1038/s41586-023-06802-1Routine clinical panels are designed to detect disease once values move outside a reference range. They say little about how fast each organ system is aging. Circulating metabolites reflect energy metabolism, detoxification, inflammation and mitochondrial function, which makes them a natural readout for organ-level aging.
I'm a PhD researcher in molecular oncology at IFOM in Milan, where I work on genome stability and how cells respond to DNA damage, two of the core processes behind both aging and cancer.
Large European biobanks have already shown that blood metabolite signatures track disease and mortality risk. That knowledge hasn't been turned into something a person in India or Europe can use to see which organ systems need attention before disease appears.
Longevity Clock is being built to close that gap: a serum metabolomics panel, calibrated for Indian and European populations, delivered as a wellness tool with a repeat-testing loop.
Five steps from sample to report. Details may change as we validate the panel.
A small serum sample, collected at a partner centre or with a home kit. Preparation instructions confirmed before launch.
80+ metabolites quantified by liquid chromatography–mass spectrometry at an accredited partner lab.
Machine-learning models (penalised regression and gradient boosting) estimate an age for each organ module.
Per-organ scores, the metabolites behind them, and where you sit against the reference cohort.
Repeat every 6–12 months to see how each organ score moves with lifestyle changes.
One person's cardiovascular markers can look young while their kidney markers look older. OLI is designed to show that map, so you know where to focus.
Bile acids, uremic toxins, ceramides, NAD⁺ intermediates, branched-chain amino acids, acylcarnitines and more.
Each metabolite group is assigned to its main organ-system pathway, with weights learned from reference data.
Initial references come from published European studies. Indian calibration will be built during the beta.
A composite model brings the six module scores together into an overall Longevity Clock age.
Primary and conjugated bile acids · hepatic lipid metabolites · amino acid detoxification markers
Uremic toxins (indoxyl sulfate, p-cresyl sulfate) · creatinine pathway · organic anion transport
Ceramides · lysophosphatidylcholines · TMAO · plasmalogens · oxidised lipid species
Tryptophan–kynurenine axis · sphingolipids · serotonin precursors · neuroinflammation markers
NAD⁺ intermediates (NMN, NAM) · acylcarnitines · TCA cycle intermediates
Branched-chain amino acids · citrate · α-ketoglutarate · insulin resistance markers
The studies that shape OLI's design, with the reason each one matters.
14 blood biomarkers predicted mortality better than conventional risk factors.
Showed organs age at different rates within a person, using blood proteins. OLI applies the same idea to metabolites.
Linked specific circulating metabolites to future cardiovascular events, informing the cardiovascular module.
Found that individuals follow different aging patterns ("ageotypes") across organ-related pathways.
Mapped blood metabolic biomarkers to over 700 diseases, a reference for module design.
Showed how strongly genetics shapes circulating metabolite levels, relevant to calibrating across populations.
Circulating signatures of exceptional longevity, useful for defining what healthy aging looks like.
These are independent studies. None of them evaluated Longevity Clock or the OLI test.
